Horseradish peroxidase (HRP) interacts with hydrogen peroxide (H₂O₂) to form an [HRP-H₂O₂] complex, which can oxidize a broad range of hydrogen donors, including various substrates used in detection. These substrates include colorless, reduced chromogenic substrates (e.g., DAB, TMB, ABTS), chemiluminescent substrates (e.g., luminol), and fluorescent substrates (e.g., Amplex Red). Upon incubation with these substrates, HRP produces a colored, luminescent, or fluorescent product, allowing quantification through colorimetric, luminescent, or fluorimetric detection methods.
HRP can be conjugated to antibodies using several techniques, such as glutaraldehyde, periodate oxidation, disulfide linkage, or amino- and thiol-directed cross-linkers.
